checkerboard correction

Has anyone ever done a checkerboard calibration to the lens/camera in order to get more accurate position location?

Like this:

Would it be possible to do this once (either using OpenCV, Matlab, or another tool) and then apply some correction factor to the pixy output on an arduino?
